Microsoft Build: 7 things to expect

Microsoft Build: 7 things to expect

Microsoft Build, which begins June 26, will be the company's first major developers' conference to be held in San Francisco since 1997. Redmond's focus on the Bay Area would have been significant under any circumstance, as the tech-heavy region is home not only to many competing conferences, such as Apple's WWDC and Google's I/O, but also to one of the densest local populations of programmers in the world. But this month's event will carry even more weight due to the company's up-and-down year, which has included triumphs such as Office 365 and Azure as well as Windows 8's controversial and underperforming debut.

At Build, Microsoft will attempt to galvanize developer interest in both its soaring and struggling platforms, and to reshape the public narrative around the divisive response to Win8. Doing so will involve a variety of efforts during the conference's three days, and though Microsoft has likely kept some surprises under wraps, industry observers have spent months predicting what Build will bring. Here are seven things Microsoft could showcase next week in San Francisco.

Two mutations triggered an evolutionary leap 500 million years ago

Two mutations triggered an evolutionary leap 500 million years ago

In a feat of "molecular time travel," the researchers resurrected and analyzed the functions of the ancestors of genes that play key roles in modern human reproduction, development, immunity and cancer. By re-creating the same DNA changes that occurred during those genes' ancient history, the team showed that two mutations set the stage for hormones like estrogen, testosterone and cortisol to take on their crucial present-day roles.

"Changes in just two letters of the genetic code in our deep evolutionary past caused a massive shift in the function of one protein and set in motion the evolution of our present-day hormonal and reproductive systems," said Joe Thornton, PhD, professor of human genetics and ecology & evolution at the University of Chicago, who led the study.

"If those two mutations had not happened, our bodies today would have to use different mechanisms to regulate pregnancy, libido, the response to stress, kidney function, inflammation, and the development of male and female characteristics at puberty," Thornton said.

The findings were published online June 24 in the Proceedings of the National Academy of Sciences.

Understanding how the genetic code of a protein determines its functions would allow biochemists to better design drugs and predict the effects of mutations on disease. Thornton said the discovery shows how evolutionary analysis of proteins' histories can advance this goal, Before the group's work, it was not previously known how the various steroid receptors in modern species distinguish estrogens from other hormones.

The team, which included researchers from the University of Oregon, Emory University and the Scripps Research Institute, studied the evolution of a family of proteins called steroid hormone receptors, which mediate the effects of hormones on reproduction, development and physiology. Without receptor proteins, these hormones cannot affect the body's cells.

Thornton's group traced how the ancestor of the entire receptor family -- which recognized only estrogens -- evolved into descendant proteins capable of recognizing other steroid hormones, such as testosterone, progesterone and the stress hormone cortisol.

To do so, the group used a gene "resurrection" strategy. They first inferred the genetic sequences of ancient receptor proteins, using computational methods to work their way back up the tree of life from a database of hundreds of present-day receptor sequences. They then biochemically synthesized these ancient DNA sequences and used molecular assays to determine the receptors' sensitivity to various hormones.

Thornton's team narrowed down the time range during which the capacity to recognize non-estrogen steroids evolved, to a period about 500 million years ago, before the dawn of vertebrate animals on Earth. They then identified the most important mutations that occurred during that interval by introducing them into the reconstructed ancestral proteins. By measuring how the mutations affected the receptor's structure and function, the team could re-create ancient molecular evolution in the laboratory.

They found that just two changes in the ancient receptor's gene sequence caused a 70,000-fold shift in preference away from estrogens toward other steroid hormones. The researchers also used biophysical techniques to identify the precise atomic-level mechanisms by which the mutations affected the protein's functions. Although only a few atoms in the protein were changed, this radically rewired the network of interactions between the receptor and the hormone, leading to a massive change in function.

"Our findings show that new molecular functions can evolve by sudden large leaps due to a few tiny changes in the genetic code," Thornton said. He pointed out that, along with the two key changes in the receptor, additional mutations, the precise effects of which are not yet known, were necessary for the full effects of hormone signaling on the body to evolve.

'Singing' rats show hope for older humans with age-related voice problems

'Singing' rats show hope for older humans with age-related voice problems

The research appears in The Journals of Gerontology.

University of Illinois speech and hearing science professor Aaron Johnson, who led the new study along with his colleagues at the University of Wisconsin, said that aging can cause the muscles of the larynx, the organ that contains the vocal folds, to atrophy. This condition, called presbyphonia, may be treatable with vocal training, he said.

Johnson said in a healthy, young larynx the vocal folds completely close and open during vibration. This creates little puffs of air we hear as sound. In people with presbyphonia, however, the atrophied vocal folds do not close properly, resulting in a gap during vocal fold vibration.

Degradation of the neuromuscular junction, or the interface between the nerve that signals the vocal muscle to work and the muscle itself, also contributes to the symptoms of presbyphonia, Johnson said. In a healthy human, when the signal reaches the neuromuscular junction, it triggers a release of chemicals that signal the muscle to contract. But an age-related decline in the neuromuscular junction can cause weakness and fatigue in the muscle, and may result in a person having a breathy or weak voice and to become fatigued as a result of the extra effort needed to communicate.

Surgery and injections may help correct the gap between the vocal folds seen in presbyphonia, but these invasive procedures are often not viable in the elderly population, Johnson said.

His previous experience working with the elderly as a former classical singer and voice teacher propelled Johnson to "become interested in what we can do as we get older to keep our voices healthy and strong."

"We know exercise strengthens the limb musculature, but we wanted to know if vocal exercise can strengthen the muscles of the voice," Johnson said.

To find out if vocal training could have an effect on the strength and physiology of the vocal muscles in humans, Johnson turned to a rat model. Rats make ultrasonic vocalizations that are above the range of human hearing, but special recording equipment and a computer that lowers the frequency of the rat calls allows humans to perceive them. (They sound a bit like bird calls).

Because rats and humans utilize similar neuromuscular mechanisms to vocalize, the rats make ideal subjects for the study of human vocal characteristics, Johnson said.

Both the treatment and control groups contained old and young male rats. In the treatment group, a female rat was placed into a cage with a male rat. When the male expressed interest in her, the female was removed from the cage, causing the male rat to vocalize. The male was rewarded with food for these vocalizations, and after eight weeks of this operant conditioning in which rewards were only given for certain responses, all of the rats in the treatment group had been trained to increase their number of vocalizations during a training session.

At the end of the eight-week period, the researchers measured the intensity of the rats' vocalizations and analyzed the animals' larynges to see whether the training had any effect on the condition of their neuromuscular junctions.

The researchers found the trained old and young rats had similar average vocal intensities, but the untrained older rats had lower average intensities than both the trained rats and the young rats that had not been trained. They also found several age-related differences within the groups' neuromuscular mechanisms.

"Other research has found that in the elderly, there is a dispersion, or breaking apart, of the neuromuscular junction at the side that is on the muscle itself," Johnson said. "We found that in the older rats that received training, it wasn't as dispersed."

These "singing rats" are the "first evidence that vocal use and vocal training can change the neuromuscular system of the larynx," Johnson said.

"While this isn't a human study, I think this tells us that we can train ourselves to use our voices and not only reduce the effects of age on the muscles of our voices, but actually improve voices that have degraded," Johnson said.

Johnson is also affiliated with the Beckman Institute for Advanced Science and Technology at Illinois.

Biological arithmetic: Plants do sums to get through the night

Biological arithmetic: Plants do sums to get through the night

"This is the first concrete example in a fundamental biological process of such a sophisticated arithmetic calculation." said mathematical modeller Professor Martin Howard from the John Innes Centre.

Plants feed themselves during the day by using energy from the sun to convert carbon dioxide into sugars and starch. Once the sun has set, they must depend on a store of starch to prevent starvation.

In research to be published in the open access journal eLife, scientists at the John Innes Centre show that plants make precise adjustments to their rate of starch consumption. These adjustments ensure that the starch store lasts until dawn even if the night comes unexpectedly early or the size of the starch store varies.

The John Innes Centre scientists show that to adjust their starch consumption so precisely they must be performing a mathematical calculation -- arithmetic division.

"The capacity to perform arithmetic calculation is vital for plant growth and productivity," said metabolic biologist Professor Alison Smith.

"Understanding how plants continue to grow in the dark could help unlock new ways to boost crop yield."

During the night, mechanisms inside the leaf measure the size of the starch store and estimate the length of time until dawn. Information about time comes from an internal clock, similar to our own body clock. The size of the starch store is then divided by the length of time until dawn to set the correct rate of starch consumption, so that, by dawn, around 95% of starch is used up.

"The calculations are precise so that plants prevent starvation but also make the most efficient use of their food," said Professor Smith.

"If the starch store is used too fast, plants will starve and stop growing during the night. If the store is used too slowly, some of it will be wasted."

The scientists used mathematical modelling to investigate how such a division calculation can be carried out inside a plant. They proposed that information about the size of the starch store and the time until dawn is encoded in the concentrations of two kinds of molecules (called S for starch and T for time). If the S molecules stimulate starch consumption, while the T molecules prevent this from happening, then the rate of starch consumption is set by the ratio of S molecules to T molecules, in other words S divided by T.

This research is funded by the Biotechnology and Biological Sciences Research Council (BBSRC)

Promising new device detects disease with drop of blood

Promising new device detects disease with drop of blood

June 24, 2013 — An NJIT research professor known for his cutting-edge work with carbon nanotubes is overseeing the manufacture of a prototype lab-on-a-chip that would someday enable a physician to detect disease or virus from just one drop of liquid, including blood. A new study describes how NJIT research professors Reginald Farrow and Alokik Kanwal, his former postdoctoral fellow, and their team have created a carbon nanotube-based device to noninvasively and quickly detect mobile single cells with the potential to maintain a high degree of spatial resolution.

"Using sensors, we created a device that will allow medical personnel to put a tiny drop of liquid on the active area of the device and measure the cells' electrical properties," said Farrow, the recipient of NJIT's highest research honor, the NJIT Board of Overseers Excellence in Research Prize and Medal. "Although we are not the only people by any means doing this kind of work, what we think is unique is how we measure the electrical properties or patterns of cells and how those properties differ between cell types."

In the article, the NJIT researchers evaluated three different types of cells using three different electrical probes. "It was an exploratory study and we don't want to say that we have a signature," Farrow added. "What we do say here is that these cells differ based on electrical properties. Establishing a signature, however, will take time, although we know that the distribution of electrical charges in a healthy cell changes markedly when it becomes sick."

This research was originally funded by the military as a means to identify biological warfare agents. However, Farrow believes that usage can go much further and potentially detect viruses, bacteria, even cancer. The research may also someday even assess the health of good cells, such as brain neurons. Since 2010, three U.S. patents, "Method of forming nanotube vertical field effect transistor," #7,736,979 (2010); "Nanotube device and method of fabrication" #7,964,143 (2011); "Nanotube device and method of fabrication" #8,257,566 (2012) were awarded for this device. In addition, more patents have been filed.

The device (shown in photo) utilizes standard complementary metal oxide semiconductor (CMOS) technologies for fabrication, allowing it to be easily scalable (down to a few nanometers). Nanotubes are deposited using electrophoresis after fabrication in order to maintain CMOS compatibility.

The devices are spaced by six microns which is the same size or smaller than a single cell. To demonstrate its capability to detect cells, the researchers performed impedance spectroscopy on mobile human embryonic kidney (HEK) cells, neurons from mice, and yeast cells. Measurements were performed with and without cells and with and without nanotubes. Nanotubes were found to be crucial to successfully detect the presence of cells.

Carbon nanotubes are very strong, electrically conductive structures a single nanometer in diameter. That's one-billionth of a meter, or approximately ten hydrogen atoms in a row. Farrow's breakthrough is a controlled method for firmly bonding one of these submicroscopic, crystalline electrical wires to a specific location on a substrate. His method also introduces the option of simultaneously bonding an array of millions of nanotubes and efficiently manufacturing many devices at the same time.

Being able to position single carbon nanotubes that have specific properties opens the door to further significant advances. Other possibilities include an artificial pancreas, three-dimensional electronic circuits and nanoscale fuel cells with unparalleled energy density.

Oddest couple share 250-million-year-old burrow

Oddest couple share 250-million-year-old burrow

The burrow revealed two unrelated vertebrate animals nestled together and fossilised after being trapped by a flash flood event. Facing harsh climatic conditions subsequent to the Permo-Triassic (P-T) mass extinction, the amphibian Broomistega and the mammal forerunner Thrinaxodon cohabited in a burrow.

Scanning shows that the amphibian, which was suffering from broken ribs, crawled into a sleeping mammal's shelter for protection. This research suggests that short periods of dormancy, called aestivation, in addition to burrowing behaviour, may have been a crucial adaptation that allowed mammal ancestors to survive the P-T extinction.

The international team of scientists was led by Dr Vincent Fernandez from Wits University, South Africa and the European Synchrotron Radiation Facility (ESRF) in Grenoble, France. The other authors from Wits University include Prof. Bruce Rubidge (Director of the newly formed Palaeosciences Centre of Excellence at Wits), Dr Fernando Abdala and Dr Kristian Carlson. Other authors include Dr Della Collins Cook (Indiana University); Dr Adam Yates (Museum of Central Australia) and Dr. Paul Tafforeau (ESRF).

After many impressive results obtained on fossils, synchrotron imaging has led to revived interest in the studies of the numerous fossilised burrows discovered in the Karoo Basin of South Africa and dated to 250 million years ago. The first attempt to investigate one of these burrow-casts surprisingly revealed a world-first association of two unrelated animals.

The fossil was recovered from sedimentary rock strata in the Karoo Basin. It dates from 250 million years ago, at the beginning of the Triassic Period. At that time, the ecosystem was recovering from the Permo-Triassic mass extinction that wiped out most of life on Earth. In the Pangea Supercontinent context, what is now South Africa was an enclave in the southern half called Gondwana. It was the scene of pronounced climatic warming and increased seasonality marked by monsoonal rainfall. To survive this harsh environment, many animals, including mammal-like reptiles (mammal forerunners), developed a digging behaviour, attested by the numerous fossilised burrow casts discovered in the Karoo Basin. These casts have long been thought to enclose fossilised remains, triggering interest from palaeontologists. Early this year, an international group of scientists started to research the contents of these burrows using X-ray synchrotron computed microtomography.

Two burrow casts were selected from the collection at Wits to be scanned using the state-of-the-art facility at the European Synchrotron Radiation Facility (ESRF). Using the unique properties of the X-ray beam which enables non-destructive probing, the scan of the first burrow started to reveal the skull of a mammal-like reptile called Thrinaxodon, an animal previously reported in another burrow.

As the scan progressed, the three-dimensional reconstruction displayed results beyond expectations: the mammal-like reptile was accompanied by an amphibian Broomistega, belonging to the extinct group of Temnospondyl.

"While discovering the results we were amazed by the quality of the images," says lead author Fernandez, "but the real excitement came when we discovered a second set of teeth completely different from that of the mammal-like reptile. It was really something else."

Besides the pristine preservation of the two skeletons, the team focused on the reasons explaining such an unusual co-habitation. Fernandez explains: "Burrow-sharing by different species exists in the modern world, but it corresponds to a specific pattern. For example, a small visitor is not going to disturb the host. A large visitor can be accepted by the host if it provides some help, like predator vigilance. But neither of these patterns corresponds to what we have discovered in this fossilised burrow."

The scientists gathered all the information to try to reconstitute the events that led to this incredible fossil aggregation, testing scenarios one after another. "It's a fascinating scientific question: what caused the association of these two organisms in the burrow? One of the more obvious possibilities is a predator-prey interaction, but we inspected both skeletons looking for tooth marks or other evidence implying predation, ultimately finding no support for one having attempted to feed on the other," says Carlson.

His colleague, Cook, adds that the consecutive broken ribs resulted from a single, massive trauma. The amphibian clearly survived the injury for some time because the fractures were healing, but it was surely quite handicapped. According to Fernandez this Broomistega is the first complete skeleton of this rare species that has been discovered. "It tells us that this individual was a juvenile and mostly aquatic at that time of its life," he says.

The scientists eventually concluded that the amphibian crawled into the burrow in response to its poor physical condition but was not evicted by the mammal-like reptile.

Numerous Thrinaxodon specimens have been found in South Africa, many of them fossilised in a curled-up position. Abdala says: "I have always been fascinated by the preservation of Thrinaxodon fossils in a curled-up position that show even tiny bones of the skeleton preserved. It's as if they were peacefully resting in shelters at the time of death."

The shelters prevented disturbance of the skeletal remains from scavengers and weathering. "We also think it might reflect a state of torpor called aestivation in response to aridity and absence of food resources," Abdala says.

Piecing all the clues together, the team finally elucidated the enigmatic association, concluding that "the mammal-like reptile, Thrinaxodon, was most probably aestivating in its burrow, a key adaptation response together with a burrowing behaviour which enabled our distant ancestors to survive the most dramatic mass extinction event. This state of torpor explains why the amphibian was not chased out of the burrow," says Rubidge.

Both animals were finally entrapped in the burrow by a sudden flood and preserved together in the sediments for 250 million years.

Tafforeau says: "Thanks to the unique possibilities for high quality imaging of fossils developed during the last decade at the ESRF, these unique specimens remain untouched, protected by their mineral matrix. Who knows what kind of information we'll be able to obtain from them in the future and which would have been completely lost if the specimen had been prepared out of its burrow cast?"

Intel joins A4WP wireless charging group

LONDON – Intel has joined the Alliance for Wireless Power (A4WP) consortium and taken a seat on the board of directors along with Broadcom, Gill Industries, IDT, Qualcomm, Samsung Electronics and Samsung Electro-Mechanics.

The move will put weight behind A4WP which is a rival standards organization to the Wireless Power Consortium (WPC), seeking to provide wireless charging for portable consumer electronics devices, including, smartphones, tablet, netbook and laptop computers.

WPC was the first to market and claims as many as 8.5 million products have shipped using its Qi technology, some based on chips announced last fall by Texas Instruments. Intel demonstrated its own technology.

A4WP, which specifies the use of magnetic resonance technology, is capable of simultaneous charging of multiples devices and the flexible positioning of devices and a charging platter.

"Intel believes the A4WP specification, particularly the use of near field magnetic resonance technology, can provide a compelling consumer experience and enable new usage models that make device charging almost automatic," said Navin Shenoy, general manager of the mobile client platform division at Intel, in a statement issued by A4WP.

The A4WP mission includes development of industry specifications for submission to national and international standards development organizations, management of an A4WP certification program, including consumer-recognizable certification logo and the coordination with national and international regulatory agencies regarding policy and compliance.

Altair attacks Qualcomm, Wi-Fi with low-cost LTE

Altair attacks Qualcomm, Wi-Fi with low-cost LTE

SAN JOSE, Calif. – Altair Semiconductor aims to drive LTE this year down into the penny-pinching budget of Wi-Fi for mobile devices and the Internet of Things. Its effort involves next-generation chips and new module suppliers such as Taiwan’s Quanta to go up against archrival Qualcomm and others.

“We want to drive LTE to Wi-Fi levels--today to add Wi-Fi to a product costs about $10 but for LTE it’s about $150-200” to the consumer, said Eran Eshed, co-founder and vice president of marketing at Altair. “Once LTE gets down to a $20-40 increment over Wi-Fi, we’ll see an inflection point,” he said, echoing the old marketing mantra of former Intel chief Andy Grove.

Altair is one of a handful of former WiMax startups encouraged by the initiative Intel had—and subsequently dropped--on that wireless alternative several years ago. Like rival Sequans, Altair now pushes an LTE-only agenda, but due to its heritage it lacks 3G cellular technology.

Altair aims to make its lack of 3G an asset. Eshed claims its LTE-only chip is smaller and cheaper and modules using it are smaller with a lower bill of materials because they do not need to support 3G bands and features.

3G filters and other components cost $3-$6 at the board level. 3G patent licenses cost even more--$5-$10, he estimates.

Altair is working on a next-gen LTE-only chip that has a smaller die thanks to a process shrink. It will mark the start of a family of LTE chips the company plans, segmented for low-end IoT to high-end, tablet devices supporting LTE Release 9+ features such as carrier aggregation.

Taiwan’s Quanta has already certified with Verizon a low-cost LTE module using Altair’s current chip. “We have big design wins you will see in late 2013 with modules for tablets and ultrabooks at new low price points,” Eshed said.

Archrival Qualcomm is the 800-pound gorilla in the cellular module market. Sierra Wireless, Novotel and others use its chips in modules Ershed claims have high margins that Quanta and other Altair contract manufacturers will undercut.

In addition to Qualcomm, Altair faces an array of formidable competitors. Broadcom, Marvell, Mediatek, Texas Instruments and GTC of Korea all have eyes on this market and the emerging IoT space. Getting an edge with low-cost LTE-only modules will not be easy—but the heated competition may spark some interesting design wins in the next year.

Robo-pets may contribute to quality of life for those with dementia

Robo-pets may contribute to quality of life for those with dementia

A study has found that interacting with a therapeutic robot companion made people with mid- to late-stage dementia less anxious and also had a positive influence on their quality of life.

The pilot study, a collaboration led by Professor Wendy Moyle from Griffith University, Australia and involving Northumbria University's Professor Glenda Cook and researchers from institutions in Germany, investigated the effect of interacting with PARO -- a robotic harp seal -- compared with participation in a reading group. The study built on Professor Cook's previous ethnographic work carried out in care homes in North East England.

PARO is fitted with artificial intelligence software and tactile sensors that allow it to respond to touch and sound. It can show emotions such as surprise, happiness and anger, can learn its own name and learns to respond to words that its owner uses frequently.

Eighteen participants, living in a residential aged care facility in Queensland, Australia, took part in activities with PARO for five weeks and also participated in a control reading group activity for the same period. Following both trial periods the impact was assessed, using recognised clinical dementia measurements, for how the activities had influenced the participants' quality of life, tendency to wander, level of apathy, levels of depression and anxiety ratings.

The findings indicated that the robots had a positive, clinically meaningful influence on quality of life, increased levels of pleasure and also reduced displays of anxiety.

Research has already shown that interaction with animals can have a beneficial effect on older adults, increasing their social behaviour and verbal interaction and decreasing feelings of loneliness. However, the presence of animals in residential care home settings can place residents at risk of infection or injury and create additional duties for nursing staff.

This latest study suggests that PARO companions elicit a similar response and could potentially be used in residential settings to help reduce some of the symptoms -- such as agitation, aggression, isolation and loneliness -- of dementia.

Prof Cook, Professor of Nursing at Northumbria University, said: "Our study provides important preliminary support for the idea that robots may present a supplement to activities currently in use and could enhance the life of older adults as therapeutic companions and, in particular, for those with moderate or severe cognitive impairment.

"There is a need for further research, with a larger sample size, and an argument for investing in interventions such as PARO robots which may reduce dementia-related behaviours that make the provision of care challenging as well as costly due to increased use of staff resources and pharmaceutical treatment."

The researchers of the pilot study have identified the need to undertake a larger trial in order to increase the data available. Future studies will also compare the effect of the robot companions with live animals.

IEEE 1149.1 'JTAG' revision could save billions

IEEE 1149.1 'JTAG' revision could save billions

IEEE has released the revised IEEE 1149.1-2013 “Standard for Test Access Port and Boundary-Scan Architecture”, commonly known in the industry as “JTAG,” for “Joint Test Action Group.”

This revision is intended to dramatically lower electronics industry costs by enabling test re-use across all phases of the integrated circuit lifecycle via vendor-independent, hierarchical test languages. The revision of IEEE 1149.1, the first for the standard since 2001, allows critical domain expertise for intellectual property (IP)—how to configure a serializer/deserializer (SERDES) for loopback testing, for example—to be transferred in a computer-readable format from the IP designer to IC designers and, in turn, to designers of printed circuit boards (PCBs) and to test engineers, gradually magnifying industry cost savings along the supply chain.

The cost savings for the electronics industry that IEEE 1149.1-2013 is intended to unlock are estimated to be in the billions of dollars. IEEE 1149.1-2013 specifies a new hierarchical Procedural Definition Language (PDL)—a standard test language based on Tcl, and hierarchical extensions to the original Boundary Scan Description Language (BSDL) to describe on-chip IP test data registers.

Eight new optional IC instructions provide a foundation for configuring I/Os for board test, mitigating false failures when re-testing the IC at the board level and correlating the results back to wafer level test through an Electronic Chip ID.

Now, the IP provider can document the IP test Interface and how to operate the IP in an English-like language—just once, for all ICs. Software tools then re-target this documentation at the IC and board level for tests. In revising IEEE 1149.1, the working group focused on two things: lowering industry costs through the new PDL language and enabling test re-use over the lifecycle of an integrated circuit.

IEEE 1149.1-2013 provides critical synergy with two other important industry standards. IEEE 1149.1-2013 supports segmented on-chip test data registers that cross power domains specified by IEEE 1801-2013 “Standard for Design and Verification of Low Power Integrated Circuits”.
